- Adjustment of brood size and androgen levels in a teleost species with exclusive male parental carePublication . Ros, Albert F. H.; Fagundes, Teresa; Oliveira, Rui FilipeIn maximizing reproductive success, individuals face a trade-off between parental care for their current offspring and investing in androgen-dependent sexual traits to produce further offspring. It has thus been proposed that parental effort would suppress androgen levels. Here, we studied parental effort by manipulating brood size in the peacock blenny, Salaria pavo, a littoral fish species in which males show long periods of paternal care.We focused on the effects of brood size on female spawning rate (measured as number of eggs received in the nest by focal males) and androgen levels. We found a positive linear relationship between brood size and the number of eggs received subsequently. Accordingly, spawning rate increased for males with experimentally enlarged broods while it decreased for males in which brood size was reduced. However, over a longer time interval, brood sizes of both treatments returned to the nonmanipulated brood size, suggesting an effect of additional factors such as male quality. Brood size did not show the expected negative relationship with levels of testosterone and 11-ketotestosterone. On the contrary, testosterone levels were positively correlated with brood size. However, in agreement with the prediction, changes in the level of parental care by manipulation of brood size showed an inverse relationship with testosterone levels. As with brood size, given time, testosterone levels tended to return to nonmanipulated levels. Such changes suggest that males adjust their brood size to an individual value through androgen modulation of courtship or other traits influencing female spawning rate.
- Adult neurogenesis in the brain of the Mozambique tilapia, Oreochromis mossambicusPublication . H.Although the generation of new neurons in the adult nervous system ('adult neurogenesis') has been studied intensively in recent years, little is known about this phenomenon in non-mammalian vertebrates. Here, we examined the generation, migration, and differentiation of new neurons and glial cells in the Mozambique tilapia (Oreochromis mossambicus), a representative of one of the largest vertebrate taxonomic orders, the perciform fish. The vast majority of new cells in the brain are born in specific proliferation zones of the olfactory bulb; the dorsal and ventral telencephalon; the periventricular nucleus of the posterior tuberculum, optic tectum, and nucleus recessi lateralis of the diencephalon; and the valvula cerebelli, corpus cerebelli, and lobus caudalis of the cerebellum. As shown in the olfactory bulb and the lateral part of the valvula cerebelli, some of the young cells migrate from their site of origin to specific target areas. Labeling of mitotic cells with the thymidine analog 5-bromo-2'-deoxyuridine, combined with immunostaining against the neuron-specific marker protein Hu or against the astroglial marker glial fibrillary acidic protein demonstrated differentiation of the adult-born cells into both neurons and glia. Taken together, the present investigation supports the hypothesis that adult neurogenesis is an evolutionarily conserved vertebrate trait.
- Aggressive behaviour and energy metabolism in a cichlid fish, Oreochromis mossambicusPublication . Ros, Albert F. H.; Becker, Klaus; Oliveira, Rui FilipeWe have investigated the effect of mirror-elicited agonistic behaviour on oxygen consumption in the Mozambique tilapia, Oreochromis mossambicus (Cichlidae). Males exposed to their mirror image showed higher frequencies of both lateral display and tail-beating and escalated aggression more frequently than males exposed to a transparent glass that was used as a control for the presence of a novel object in the tank. This aggressive response was correlated with an increase in oxygen consumption. Overt aggression was highly correlated with display behaviour and with locomotor activity. Bivariate analyses showed high correlation (explaining about 64% of variation) between overt aggression, locomotor activity and metabolic rates. Weakly positive bivariate correlations between displays and metabolic rates turned spurious after partialling out aggression. The data suggest that energetic costs only emerge late during the conflict, when animals escalate their aggressive behaviour.
- Agonistic interactions elicit rapid changes in brain nonapeptide levels in zebrafishPublication . Teles, Magda; Gozdowska, Magdalena; Kalamarz-Kubiak, Hanna; Kulczykowska, Ewa; Oliveira, Rui FilipeThe teleost fish nonapeptides, arginine vasotocin (AVT) and isotocin (IT), have been implicated in the regulation of social behavior. These peptides are expected to be involved in acute and transient changes in social context, in order to be efficient in modulating the expression of social behavior according to changes in the social environment. Here we tested the hypothesis that short-term social interactions are related to changes in the level of both nonapeptides across different brain regions. For this purpose we exposed male zebrafish to two types of social interactions: (1) real opponent interactions, from which a Winner and a Loser emerged; and (2) mirror-elicited interactions, that produced individuals that did not experience a change in social status despite expressing similar levels of aggressive behavior to those of participants in real-opponent fights. Non-interacting individuals were used as a reference group. Each social phenotype (i.e. Winners, Losers, Mirror-fighters) presented a specific brain profile of nonapeptides when compared to the reference group. Moreover, the comparison between the different social phenotypes allowed to address the specific aspects of the interaction (e.g. assessment of opponent aggressive behavior vs. self-assessment of expressed aggressive behavior) that are linked with neuropeptide responses. Overall, agonistic interactions seem to be more associated with the changes in brain AVT than IT, which highlights the preferential role of AVT in the regulation of aggressive behavior already described for other species.
- Alternative male reproductive tactics and the immunocompetence handicap in the azorean rock-pool blenny, Parablennius parvicornisPublication . Ros, Albert F. H.; Bouton, Niels; Santos, Ricardo Serrão; Oliveira, Rui FilipeIn the Azorean rock-pool blenny (Parablennius parvicornis) reproductively active males display alternative morphotypes, which differ in the expression of secondary sexual characters (SSC). Males expressing SSC, theMCmorphotype, have high androgen levels and compete for crevices that will be visited by females to spawn.MCmales holding nests court females and care for the eggs. Males with low expression of SSC, the MK morphotype, have low levels of androgens and reproduce by stealing fertilizations from the MC males. Based on the hypothesis that androgens are immunosuppressive, we expected these morphotypes to differ in immunocompetence. To test this hypothesis, we conducted a field study in which we collected repeated blood samples to monitor leukocyte populations (blood smears), and to measure the primary antibody response of males that were experimentally challenged with a foreign non-pathogenic antigen (sheep red blood cells). Circulating levels of 11-ketotestosterone and testosterone were higher in MC males than in MK males. Neither granulocyte nor thrombocyte counts did covariate with androgens or male tactic. In contrast, lymphocyte counts and humoral antibody response were negatively correlated with body size, and as expected, both were lower in MC than in MK males. Interestingly, in MC males androgen levels decreased after immunization, and this was less in nest-holder males than in MC males that were floating around in the pools. Within each morphotype we found no relationship between androgens and immunocompetence. The latter result is not supportive for androgen regulated immunosuppression in MC males. A possible alternative is enhancement of immunity in MK males. These males had relatively high levels of injuries in comparison withMCmales. High immunity might be a consequence of high infection rate because of such injuries.

M.Nest-holder male Salaria pavo have lower circulating concentrations of 11-ketotestosterone (KT) at the beginning of the breeding season than at its peak. At that peak density of nesting males was higher as were the number of visits of sneaker males to nests and of agonistic interactions between nest-holders and sneaker males. There was no difference between the two dates either in the frequency of male–male interactions or in the frequency of courtship episodes. Thus, higher plasma levels in nest-holders might be explained by a more intense sneaking pressure at the peak of the breeding season. At that peak, nest-holders had higher plasma levels of KT and a higher testosterone (T) to KT metabolization index in the gonads than did floater males. Both nest-holders and floaters had higher levels of KT and T in the testicular gland than in the testis. The levels of both androgens in the testicular gland, but not in the testis, were correlated with circulating concentrations of KT. These results suggest that the testicular gland is the major source of circulating KT in blenniids. Nest-holders had higher metabolization indexes than floaters both in the testis and in the testicular gland, which suggests that nest holding status promotes the conversion of T into KT.
- Androgen modulation of social decision-making mechanisms in the brain : An integrative and embodied perspectivePublication . Oliveira, Gonçalo A.; Oliveira, Rui FilipeApart from their role in reproduction androgens also respond to social challenges and this response has been seen as a way to regulate the expression of behavior according to the perceived social environment (Challenge hypothesis, Wingfield et al., 1990). This hypothesis implies that social decision-making mechanisms localized in the central nervous system (CNS) are open to the influence of peripheral hormones that ultimately are under the control of the CNS through the hypothalamic-pituitary-gonadal axis. Therefore, two puzzling questions emerge at two different levels of biological analysis: (1) Why does the brain, which perceives the social environment and regulates androgen production in the gonad, need feedback information from the gonad to adjust its social decision-making processes? (2) How does the brain regulate gonadal androgen responses to social challenges and how do these feedback into the brain? In this paper, we will address these two questions using the integrative approach proposed by Niko Tinbergen, who proposed that a full understanding of behavior requires its analysis at both proximate (physiology, ontogeny) and ultimate (ecology, evolution) levels.
- Androgen response to social competition in a shoaling fishPublication . Teles, Magda; Oliveira, Rui FilipeAndrogens respond to social challenges and this response has been interpreted as a way for males to adjust androgen-dependent behavior to social context. However, the androgen responsiveness to social challenges varies across species and a conceptual framework has been developed to explain this variation according to differences in the mating system and parental care type, which determines the regimen of challenges males are exposed to, and concomitantly the scope (defined as the difference between the physiological maximum and the baseline levels) of response to a social challenge. However, this framework has been focused on territorial species and no clear predictions have been made to gregarious species (e.g. shoaling fish), which although tolerating same-sex individuals may also exhibit intra-sexual competition. In this paper we extend the scope of this conceptual framework to shoaling fish by studying the endocrine response of zebrafish (Danio rerio) to social challenges. Male zebrafish exposed to real opponent agonistic interactions exhibited an increase in androgen levels (11-ketotestosterone both in Winners and Losers and testosterone in Losers). This response was absent in Mirror-fighters, that expressed similar levels of aggressive behavior to those of winners, suggesting that this response is not a mere reflex of heightened aggressive motivation. Cortisol levels were also measured and indicated an activation of the hypothalamic-pituitary-interrenal axis in Winners of real opponent fighters, but not Losers or in Mirror-fighters. These results confirm that gregarious species also exhibit an endocrine response to an acute social challenge.
- Androgenization of dominant males in a cichlid fish: Androgens mediate the social modulation of sexually dimorphic traitsPublication . Oliveira, Rui Filipe; Almada, Vítor CarvalhoWe present data on the relationship between social status and male secondary sex traits, both morphological and ethological, in Oreochromis mossambicus at the onset of sexual maturation. There is a differential expression of morphological (dorsal fin height, anal fin height, mandible width and premaxilla length) and behavioural (nuptial coloration, per cent time defending a territory, spawning pit volume, pit hovering and courtship rate) traits according to social status, with dominants presenting the highest expression of these dimorphic traits. Social status also affects the development of the genital papillae, which is considered to be a good predictor of androgen levels in cichlid fishes. The results support a causal model in which social status modulate androgen levels that in turn modulate the expression of behavioural and morphological male traits.
- Androgens and immune function in male alternative reproductive morphotypes of the peacock blenny Salaria pavoPublication . Ros, Albert F. H.; Oliveira, Rui FilipeSpecies with alternative reproductive tactics offer the opportunity to analyse how behavioural and morphological traits are tuned to produce successfully competing phenotypes within one sex. In the teleost fish Salaria pavo, alternative reproductive tactics are sequential. The older ornamented males compete for access to females by guarding a cavity to which they attract females to spawn. Ornamented males that are found without a nest are called ‘floaters’. Younger mature males which are too small to compete with nest-holders attempt to ‘sneak’ as female mimics into successful nests and release their sperm to fertilize freshly spawned eggs. The alternative tactics in S. pavo are associated with different levels of the androgens testosterone and 11-ketotestosterone, which have been found to suppress immune function in several teleost fish. A field study was carried out to analyse the relationship between these reproductive tactics, androgens and blood levels of lymphocytes as a monitoring method of immune function. We expected highest investment in the immune system in sneakers because these have the lowest androgen levels and functionally because investing in self-maintenance increases their future prospect to switch tactic and to reproduce as a nest-holder. Indeed, the relative count of lymphocytes correlated negatively with body size and thus was highest in sneakers and lowest in nest-holders. In concordance, 11-ketotestosterone and testosterone levels were found to be higher in floaters and nest-holders than in sneakers. However, no correlation was found between individual levels of testosterone or 11-ketotestosterone and lymphocytes. Thus, a trade-off between reproductive traits associated with high androgen levels and immunocompetence might exist at the level of alternative tactics but this might not be explained by acute immunosuppressive effects of circulating androgens.
